Progress in spatial localization of single cells in microenvironment at our college

In the field of single-cell analysis, electrical impedance measurement has been widely applied in single-cell characterization as a non-destructive and low-cost method. However, uncertainty in the spatial location of cells in microfluidic channels can cause impedance measurement errors. In order to eliminate the effect of spatial position, it is necessary to accurately locate the spatial position of single cells. Associate Professor Huang Liang's group at the School of Instrumentation Science and Optoelectronic Engineering, Hefei University of Technology, proposed a single-cell spatial position measurement method based on floating electrodes, which realizes the accurate measurement of three-dimensional spatial localization and velocity information of single cells, and achieves a positioning accuracy of 2.1 μm and 1.2 μm in the horizontal and vertical position, respectively. This work provides an effective solution for correcting the bias of single-cell electrical impedance measurement.

The related research paper Floating-Electrode-Enabled Impedance Cytometry for Single-Cell 3D Localization has been published in the journal Analytical Chemistry, and was selected as the cover of the 95th issue. In addition, the group participated in the 26th International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2022), which was organized by the Chemical and Biological Microsystems Society (CBMS), and won the Best Academic Poster Award.

The first author of the paper is Qiang Fang, a master student of Hefei University of Technology, and the corresponding author is Associate professor Liang Huang. The research were supported by the National Natural Science Foundation of China and the Natural Science Foundation of Anhui Province, with Tsinghua University as co-author.
